2. DAPI staininga. Media를 제거한 후 1x PBS로 3번 washing
b. Fixing : 3.7% formaldehyde 1ml을 well에 넣은 후 상온에서 15분간 둔다.
c. 1x PBS로 3번 washing 한다.
d. Slide glass 위에 DAPI mounting solution을 한 방울 떨어뜨린다.
e. Cell이 붙어있는 cover glass를 mounting solution 위에 조심스럽게 올려놓는다.
f. 건조 시킨 후 형광 현미경으로 관찰한다.
3. DNA fragmentation
a. Media를 제거한 후 1x PBS로 3번 washing한다.
b. 1ml 1x PBS를 well에 넣고 scraper로 cell을 harvest 한다.
c. EP-tube에 담고 6000rpm으로 5분간 centrifugation 한 후, Supernatant 제거
d. Lysis buffer(RIPA buffer) 100㎕를 넣고 suspension 후 얼음에 10분간 둔다.
e. RNase A 1㎕를 넣고 37℃에서 30분간 반응시킨다.
f. Proteinase K 1㎕를 넣고 45℃에서 2시간 반응시킨다.
g. PCI를 100㎕(1:1 volume)을 넣고 voltexing
PCI = Phenol:Chloroform:Isoamylalcohol = 25:24:1
h. 13000rpm으로 10분간 centrifugation(at RT) 후, Sup(약 80㎕)을 새 EP-tube로 옮긴다.
i. NaAc 10㎕(1/10 volume), glycogen 1㎕, 100% EtOH 250㎕(2.5volume)을 넣고
inverting. 이때 최종 EtOH 농도는 70%
j. -70℃에서 15분 둔 후, 13000rpm 4℃로 10분간 centrifugation
k. Sup을 버린 후 70% EtOH 1ml 넣고 13000rpm 4℃로 10분간 centrifugation
l. Sup을 완전히 따낸 후, air dry (gel에 sample을 넣을 때 buffer에 뜨는 것을 방지)
m. gel(0.8%) electrophoresis를 통하여 분석.
