1. What major features are included in the TOPO vector?
TOPO vector carries four critical elements:
Selection marker genes: lac Z & antibiotic resistance genes
Lac Z gene encodes lactose-degrading enzyme called β-galactosidase. This is essential in screening blue-white colonies for the presence of the DNA insert. In addition, functional β-galactosidase is established from two polypeptides known as lac Z α-fragment & Ω-fragment.
<Results/Discussion>
Several observations and remarks are made according to the results shown above. First of all, the control group on the LB agar plate (+ampicillin) will obviously not form any colonies as illustrated in figure 1. This is because competent E.coli without transformation is not resistant against ampicillin due to lack of either recombinant or normal TOPO plasmid and hence are compromised by the antibiotic. Therefore, no growth or colonies are formed.
