This experiment is measurement of Hill activity using isolated chloroplast. There are photosynthesis in plants. This process is two steps; Light reaction and Dark reaction. In this time, we observe the effect of light intensity on Hill reaction activity of isolated chloroplasts particles and herbicides(DCMU) on this activity. We use dichlorophenol indophenol(DCPIP) as the electron acceptor of photosystem H activity. Oxidized DCPIP is blue while reduced DCPIP is colorless. So we can observe change of color and know the progress of the reduction. As a result, as time goes on, solution is colorless. And we use DCMU also. This reagent blocks electron transport to DCPIP. So we can observe blue color.
